Formal, Samuel B., (Walter Reed Army Institute of Research, Washington, D.C.), E. H. LaBrec, T. H. Kent, and S. Falkow. Abortive intestinal infection with an Escherichia coli-Shigella flexneri hybrid strain. J. Bacteriol. 89:1374-1382. 1965.-The mechanism of the apparent loss of virulence of an Escherichia coli-Shigella flexneri hybrid strain was studied. The parent Shigella strain caused a fatal enteric infection when fed to starved guinea pigs, and signs of dysentery followed its oral administration to monkeys. The hybrid strain failed to produce any apparent symptoms when fed to either of these species. The parent strain was shown to invade the intestinal mucosa of starved guinea pigs. This caused a severe inflammatory reaction in the lamina propria, which progressed to ulceration of the intestinal epithelium and resulted in death of the animal. The hybrid strain also invaded the intestinal mucosa and produced an inflammatory reaction. In this case, the inflammatory reaction subsided, the intestine returned to normal within 4 days after challenge, and the animal survived. Both fluorescent-antibody techniques and in vivo growth studies have shown that the hybrid strain can not maintain itself in the intestinal mucosa. Preliminary studies have indicated that a similar situation also exists in the monkey. It is concluded that the virulence of dysentery bacilli rests not only in the capacity to reach the lamina propria, but also in the ability to multiply in this region.

Factors affecting virulence of Shigella flexneri: strain with relaxed control of ribonucleic acid synthesis.

An Escherichia coli-Shigella flexneri hybrid strain which produces an abortive, nonfatal experimental dysentery has inherited relaxed control of ribonucleic acid synthesis from its E. coli parent. This property, coupled with a partial methionine requirement, has been used to explain its long lag phase in vitro, relative to its Shigella parent. It was suggested that such characteristics may cont...

Mechanisms and genetics of resistance to sodium lauryl sulfate in strains of Shigella and Escherichia coli.

The sensitivity to sodium lauryl sulfate (SLS) of Shigella flexneri and Escherichia coli is determined by at least three genes. One site is located near the lactose operon, and two loci are cotransducible with the arabinose operon. Calcium ions protect against SLS lysis. One gene is concerned with the relative ability of the bacterium to retain calcium against such chelating agents as ethylened...

Persistence of an Escherichia coli-Shigella flexneri hybrid in the intestinal tract of Macaca mulatta.

A live oral vaccine prepared from an Escherichia coli-Shigella flexneri 4b hybrid was administered by gavage to Macaca mulatta. In the three main studies involving 160 monkeys, three doses were used, generally consisting of 5 x 10(9) to 60 x 10(9) cells per dose, with an interval between doses of 6, 7, or 14 days. Rectal swab cultures at the time of the last vaccine dose, and 14 to 58 days late...

EspG, a novel type III system-secreted protein from enteropathogenic Escherichia coli with similarities to VirA of Shigella flexneri.

The function of the rorf2 gene located on the locus of enterocyte effacement (LEE) pathogenicity island of enteropathogenic Escherichia coli (EPEC) has not been described. We report that rorf2 encodes a novel protein, named EspG, which is secreted by the type III secretory system and which is translocated into host epithelial cells. EspG is homologous with Shigella flexneri protein VirA, and th...

Interaction between Escherichia coli and Shigella flexneri in the digestive tract of "gnotobiotic" mice.

When a strain of Shigella flexneri and then a strain of Escherichia coli were implanted at an interval of 1 day in the digestive tract of germfree mice, the S. flexneri was eliminated in 8 days. However, when S. flexneri was mono-associated with germfree mice in the absence of E. coli, a population of S. flexneri appeared which was resistant to the antagonistic effects of E. coli in vivo. The e...
